The biosynthesis of radioactive fatty acids and cholesterol.

چکیده
The biological synthesis of long chain fatty acids from acetate by slices of rat liver tissue was demonstrated by Bloch, Borek, and Rittenberg (1) in 1946, and has been repeated by several investigators (2, 3). The effect of insulin on this incorporation was studied by Bloch and Kramer (2), who reported that, whereas insulin inhibited the incorporation of labeled acetate into fatty acids, the combination of insulin and non-labeled pyruvate resulted in an increased incorporation of the labeled acetate. It was of interest to determine whether other short chain fatty acids, as well as acetic acid, might be utilized by liver slices for the synthesis of higher fatty acids. This report isaccordingly concerned with the utilization of carboxyl and methyl-C4-acetic acid, /3-C14-pyruvic acid, and carboxylC4-butyric, hexanoic, and octanoic acids for this biosynthesis. The experiments reported here reveal some information concerning the mechanism by which these substrates are incorporated into fatty acids. A study of the effect of insulin upon this process has also been made.

Background: For understanding the roles of fatty acids on the induction of acrosome reaction which occurs under association of cholesterol efflux and PKA or PKC pathways in boar spermatozoa, metabolic fate of alone and combined radiolabeled 14C-oleic acid and 3H-linoleic acid incorporated in the sperm was compared, and behavior of cholesterol and effects of PKA and PKC inhibitors upon fatty aci...

متن کاملType of Fatty Acid Intake, Serum Lipid and Lipoprotein Levels in relation to Creatinine and GFR
Correlation between fatty acids intake, serum lipid and lipoproteins with serum creatinine and kidney function in patients with renal failure has been suggested by other investigators. This relationship has not been reported for all types of fatty acids. This study was conducted to determine the association of the consumption of different type of fatty acids, serum lipids and lipoproteins with ...
متن کاملThe synthesis of radioactive cholesterol and fatty acids in vitro.
It has been established that rat liver slices are able to convert acetate (l), acetone (2), pyruvate (3, 4), butyrate, hexanoate, and octanoate (4) to cholesterol and long chain fatty acids. Zabin and Bloch (5) and Price and Rittenberg (6) administered 1 ,3-C14-acetone to rats and demonstrated that it is converted to fatty acids and cholesterol. The former investigators (5) also reported that m...
